Pep-Optima

It's almost impractical to screen the combinatorial library of an oligopeptide of >=6 aa residues. Pep-Optima can simultaneously synthesize a combinatorial library of two AA sites on 400 spots of cellulose membrane (figure a), enabling scientistes high-throughput screening peptides in a time- and cost-efficient manner.

To optimize the sequence of all amino acid redidues in a peptide, a more comprehensive scan will be employed. The scan will begin with a combinatorial library of 2 amino acid residues (in other words, only two sites are defined and other aa residues are randomized by coupling with a mixture of 20 AAs), followed by subsequent deconvolution libraries. For example, to screen the best AA sequence of hexapeptide, a combinatorial library of 2 amino acid residues (X-X-B1-B2-X-X, where B1 and B2 are substituted by 20 amino acids, and X represents random aa residue by using a mixture of 20 aa) will be first synthesized on 400 spots array (each spot has a sub-library of 204 peptides) and screened for the best B1B2 combination (figure b). Then, the repeated round will identify the best aa combination of next two AA residues, and three rounds in total are able to obtain the best aa sequence of a hexapeptide from a potential library of 206 peptides.

Figure a. for a given peptide of D-V-L-A-F-X1-X2-Q-R-P-V-T, where X1 and X2 are replaced with any 19 amino acids other than cysteine (to avoid disulfide bond between two cysteine), there are 361 combinations (peptides). Pep-Optima can synthesize all 361 peptides in parallel on cellulose membrane for high-throughput screening.

  • Calculation of total amino acid residues: for a simple combination of AAs at two sites of a hexamer, there are 20×20=400 combinations (peptides) , the number of total amino acid residues is 400×6=2400. For comprehensive combination of a hexapeptide, 3 round of simple combinations are required (4 rounds for a octapeptide, 5 rounds for a decapeptide, and 400 sub-libraries for each round); therefore, the number of total AAs are 2400×3=7200.
