Pep-Overlap
In Pep-Overlap arrays, peptides are derived from and walk downstreams along the aa sequence of a target protein and two consecutive/neighboring peptides have several aa overlap (1~3 aa shift/offset is usually recommended for fine-mapping), and these peptides are also called overlapping peptides.
By incubating Pep-Overlap array with bait proteins (such as serum antibodies, pure and partial purified proteins, and other biomolecules), the (sub-)domains (such as epitopes of an antigen, binding motifs for a bait protein, etc) in the target protein can be quickly identified.

Applications:
- Epitope Mapping: Antigens are laid in the form of overlapping peptides on peptide array which can be incubated with sera, and then detected by secondary antibody. Useful for antibody profiling, sub-unit vaccine development.
- immune monitoring of vaccine responses
- Mapping of binding motifs for protein-protein interaction (substrate-enzyme, ligand-receptor, etc): Target protein is laid on peptide array, incubated with bait protein and then detected by the antibody against bait protein; vice versa for finding binding motifs on bait proteins.
- Binding motifs of a proteins for binding with other biomolecules: Protein-nucleic acid, protein-extracellular matric etc. Other biomolecules must be detected by either direct labeling (e.g. biotin, fluorochrome, isotopes etc) or indirect detection (e.g. antibodies, etc).
